Description
  • Microsatellites are the polymorphic regions of DNA. They are inherited genetically strictly according to Mendelian laws, so they are very powerful markers to study extra pair copulations, population genetics, ecology or migration of animals. The search for the new microsatellite loci from a ´new´ species usually requires the cloning and sequencing of their DNA. An alternative way is a cross-species amplification of their genomic DNA with primers, which are known in other species usually from the same class. We collected blood samples of 116 adult individuals (males and females) caught in the Jeseníky Mountains, Northern Moravia, the Czech republic in three years (1999, 2000 and 2001), in the Krkonoše Mountains in the north of the Czech republic (2001 and 2002), and in the Tydal province in central Norway (1998 and 2000). The DNA from blood in the storage buffer was isolated using the phenol - chloroform method. 34 primer pairs were used for the PCR amplification; these primer pairs were used to study t

Tito ptáci byli odchyceni v Jeseníkách v letech 1999, 2000 a 2001, v Krkonoších v letech 2001 a 2001 a ve středním Norsku v provincii Tydal v letech 1998 a 2000. DNA byla z krve uložené v konzervačním pufru vyizolovány fenol-chloroformovou metodou. PCR amplifikací bylo odzkoušeno celkem 34 párů primerů, které se používají pro amplifikaci mikrosatelitových lokusů v ptačí DNA: Lox1, Lox2, Lox3, Lox6, Lox7, Lox8, Ase10, Ase13, Ase16, Ase18, Ase21, Ase25, Ase34, Ase3 (cs)
