  • Objectives: Little is known about the effect of adipokines in the pathogenesis of the acute myocardial infarction (AMI) in young patients. The aim of this pilot study was to evaluate whether it is possible to detect mRNA expressions of selected adipokines in the peripheral blood leukocytes of young patients with myocardial infarction. Methods: We prospectively examined 42 patients who had an acute myocardial infarction (AMI) at a young age (men 18-45 years of age and women 18-55 years of age) and control groups of 31 patients with metabolic syndrome and 15 healthy individuals. RNA was isolated from their peripheral blood leukocytes and transcribed into cDNA (transcription kit iSCRIPT). Subsequently, the expression of the following genes were analysed (LC 480II, BioVendor kits): Adipocyte Fatty Acid Binding protein (AFABP, FABP4), Leptin and Adiponectin receptor 2 (ADN-R2). Beta-2-microglobulin was examined as a housekeeping gene. Results: In the group of patients after AMI the expression of ADN-R2 was detected in 80% patients, Leptin in 75% and FABP4 expression in 67.5%. In the control group AND-R2 and FABP4 gene were expressed in all subjects and Leptin expression was absent in 2 healthy subjects (4.3%). A correlation could not be found between FABP4 gene expression and the concentration of its product (AFABP) in the serum of the group with AMI. Conclusion: We have shown that the mRNA expression of selected adipokines can be quantified in the peripheral blood leukocytes. Whether low expressions of protective adipokines contribute to early manifestations of acute myocardial infarction needs to be explored in further studies.
