Description
  • The software is designed for microscopic stations equipped by automatically controlled table using the (x,y,z) coordinate system. First, the software generates coordinates for the pre-scan acquisition involving the whole sample. This is done in the low resolution mode using 4x or 10x objective. This step generates a set of preview images. These images are displayed sequentially by the software and the user can use simple graphical tools for manual identification of the centers of objects of interest. Based on these positions the software automatically generates new set of (x,y,z) coordinates for the automatic microscopic table allowing further acquisition in detailed resolution (e.g. using 20x,40x, 63x and 100x objectives). The software also enables recalibration of the sample in case the sample has to be manipulated between the pre-scan and the high resolution scanning (e.g. to supply the immersion oil). Other extra function of the software is recalibration of the z-coordinates for all the objects of interest using interpolation of four automatically selected objects of interests which introduce the outermost corners of the sample. This function is particularly important in case the sample is not exactly straight to significantly reduce the demands on the focus function.
