| - CÍLE: Ibotenová kyselina, muscimol a muskarin jsou zodpovědné za psychotropní efekt muchomůrky červené a tygrované. Potřeba jejich specifické a citlivé identifikace a stanovení v biologickém materiálu vedla ke snaze vyvinout spolehlivou analytickou metodu, ale uspokojivá metoda však zatím chybí. Předkládaný článek popisuje kapalinovou chromatografii s hmotnostní detekcí jako vhodnou metodu izolace a identifikace hlavních toxinů muchomůrky červené a tygrované v moči. METODY A VÝSLEDKY: Pracovní postup se skládá z extrakce analytů na SPE kolonkách Strata X-CW a Discovery SCX a separace, které je dosaženo použitím kolony Gemini C18 (150 mm x 2.0 mm, 5 micron) a 8 mM heptafluoromáselné kyseliny jako mobilní fáze. Detekce je provedena při m/z 159 pro ibotenovou kyselinu, m/z 115 pro muscimol a m/z 174 pro muskarin. Retenční časy a LOD jsou 2.6 min a 50 ng.ml-1 pro ibotenovou kyselinu, 4.6 min a 40 ng.ml-1 pro muscimol a 14.2 min a 3 ng.ml-1 pro muskarin. ZÁVĚR: Byla vyvinuta citlivá a specifická metoda (cs)
- OBJECTIVES: Ibotenic acid, muscimol and muscarine were recognized as responsible for psychotropic effects of A. muscaria and A. pantherina. Demand for their specific and sensitive identification and quantitation in biological material lead to effort to develop reliable analytical method, but satisfactory solution is still lacking. Presented article describes liquid chromatography-mass spectrometry method suitable for isolation and identification of principal toxins of A. muscaria and A. pantherina in urine. METHODS AND RESULTS: Dedicated liquid chromatography-mass spectrometry method is reported. Technique consists of an extraction of analytes on Strata X-CW and Discovery SCX SPE cartridges and separation is achieved using a Gemini C18 column (150 mm x 2.0 mm, 5 micron) and 8 mM heptafluorobutyric acid as mobile phase.
