  • Chronická expozice kůže UV zářením představuje rizikový faktor vzniku kožních nádorů. UV světlo, zejména UVA část, vyvolává v buňkách zvýšenou produkci reaktivních kyslíkových sloučenin (ROS), které následně poškozují různé buněčné makromolekuly. Aplikace antioxidantů představuje jednu z možných strategií, jak potlačit oxidační poškození vyvolané UV zářením. V této studii byl testována schopnost flavonolignanů silybinu (SB) and 2,3-dehydrosilybinu (DS) (1-50 mmol/l), obsažených v semenech ostropestřce mariánského (Silybum marianum), snížit poškození navozené UVA světlem. Lidské keratinocyty HaCaT byly použity jako experimentální in vitro model. Účinek flavonolignanů byl sledován na úrovni buněčné viability, proliferace, intracelulární hladiny ATP a glutathionu, produkce ROS, lipoperoxidace buněčných membrán, aktivace/aktivity kaspasy 3 a DNA. Flavonolignany (1-50 mmol/l) zvyšovaly viabilitu UVA ozářených (20 J/cm2) keratinocytů. SB a DS rovněž potlačovaly vyčerpání intracelulárního ATP a glutathio (cs)
  • BACKGROUND: UV radiation from sunlight is a very potent environmental risk factor in the pathogenesis of skin cancer. Exposure to UV light, especially the UVA part, provokes the generation of reactive oxygen species (ROS), which induce oxidative stress in exposed cells. Topical application of antioxidants is a successful strategy for protecting the skin against UV-caused oxidative damage.
