| - The most effective surgical therapy for end-stage hip and knee osteoarthritis is total joint arthroplasty. On the other hand, achieved outcomes deteriorate with time mainly due to osteolysis and aseptic loosening. Both of them are associated with polyethylene wear deliberating from artificial joints during each step. - Nejefektivnější terapeutický postup při konečném stádiu osteoartrózy kyčelního nebo koleního kloubu je umělá náhrada tohoto kloubu. Jednou z nejzávažnějších komplikací je omezená životnost kloubní náhrady spojená s uvolněním implantátu z kostního lůžka vlivem periprotetické osteolýzy, vedoucí k jeho selhání. Tento jev je způsoben uvolňováním polyethylenových částic z umělého kloubu vlivem otěru při každém pohybu. Tyto otěrové částice jsou fagocytovány makrofágy, což vede ke spuštění kaskád procesů vedoucích k tvorbě otěrového granulomu, k resorpci kosti a k uvolnění implantátu. Předpokládá se, že synoviální tekutina obsahuje všechny signální molekuly podílející se na patofyzilogii procesu selhání kloubní náhrady vlivem otěrových (polyethylenolvých) částic. Cílem této studie bylo určit vliv vzorků synoviální tekutiny pacientů se selhávající nebo dobře fungující umělou náhradou kyčelního/koleního kloubu na životnost a proliferaci SaOS-2 buněčné kultury.
