Description
| - Natural compounds have been used in the treatment of various diseases for centuries. Herein, we investigated the effects of structurally diverse alkaloids with anti-inflammatory activity (berberine, sanguinarine, chelerythrine, and colchicine) on two important anti-inflammatory and pro-inflammatory players, i.e. glucocorticoid receptor (GR) and nuclear factor kappa B (NF-ęB), respectively. Sanguinarine and chelerythrine elicited nuclear translocation of the p65 subunit of NF-ęB. The nuclear import of p65 was strongly augmented by these akaloids in non-stimulated cells as well as in cells challenged with tumor necrosis factor alpha (TNFá). Colchicine and berberine had no effect on p65 nuclear translocation regardless of the presence or absence of TNFá. Colchicine caused rapid degradation of the GR protein, whereas berberine had no effect on GR content or cellular localization. - Přírodní látky byly používány v léčbě různých onemocnění po staletí. V této práci jsme studoval účinek strukturně odlišných alkaloidů s protizánětlivou aktivitou (berberinu, sanguinarinu, chelerythrinu a kolchicinu) na dva důležité protizánětlivé a prozánětlivé faktory, tj. glukokortikoidní receptor (GR) a nukleární faktor kappa B (NFkB). Sanguinarin a chelerythrin vyvolaly jadernou translokaci p65 podjednotky faktoru NFkB. Jadený import p65 byl těmito alkaloidy výrazně zesílen v nestimulovaných buňkách stejně tak jako v buňkách inkubovaných s tumor nekrotizujícím faktorem alfa (TNFa). Kolchicin a berberin neměly žádný účinek na jadernou translokaci p65 bez ohledu na přítomnost nebo nepřítomnost TNFa. Kolchicin způsobil rychlou degradaci GR proteinu, zatímco berberin neměl vliv na obsah a buněčnou lokalizaci GR.
