| - Ke studiu biosyntézy cytokininů byl zvolen a charakterizován teplotně-senzitivní mutant oveST25 mechu Physcomitrella patens, u něhož po termoindukci dochází ke změnám hladin cytokininů. Kombinací kapalinové chromatografie a hmotové spektrometrie (LC-MS) bylo zjištěno, že tepelná indukce (25oC) vede u oveST25 k výraznému zvýšení extracelulárních koncentrací N6-(Δ2-isopentenyl)adeninu (iP), N6-(Δ2-isopentenyl)adenosinu (iPR), cis-zeatinu (cZ), cis-zeatinribosidu (cZR) a jeho O-glukosidu (cZROG), zatímco u kontroly (wild-type; WT) nebyly nalezeny žádné změny. Pro stanovení významu tRNA na produkci cytokininů u Physcomitrella byly analyzovány hladiny cytokininů v pletivech, kultivačním médiu a v tRNA hydrolyzátech mechu. V extraktech WT kultury bylo zjištěno, že 56% veškerých cytokininů představují deriváty iP, 32% deriváty cZ a 11% deriváty trans-zeatinu (tZ). Také v tRNA převládají různé formy cZ (90% všech cytokininů) a iP (8%), zatímco cytokininy tZ-typu byly detekovány pouze v stopových množstv... (cs)
- To study cytokinin biosynthesis, we characterized a temperature-sensitive cytokinin-overproducing mutant, oveST25, of the moss Physcomitrella patens with respect to changes in cytokinin content during thermal induction in comparison to wild type. Our findings, based on combined liquid chromatography-mass spectrometry (LC-MS) analyses, show that thermoinduction caused a strong increase of extracellular N 6-(Δ2-isopentenyl)adenine (iP), N 6-(Δ2-isopentenyl)adenosine (iPR), cis-zeatin (cZ), cis-zeatin riboside (cZR) and its O-glucoside cZROG in oveST25. In contrast, no significant changes were measured in the wild type. To investigate the relevance of tRNA for cytokinin production in Physcomitrella, we determined cytokinins in tissue and culture medium as well as in tRNA hydrolysates. The analysis of cytokinins from whole-culture extracts of wild type revealed 56% of iP-type, 32% of cZ-type, and 11% of trans-zeatin (tZ)-type forms.
