  • Flow cytometry (FCM) is extensively used to estimate DNA ploidy and genome size in plants. In order to determine nuclear DNA content, nuclei in suspension are stained by a DNA-specific fluorochrome and fluorescence emission is quantified. Recent studies have shown that cytosolic compounds may interfere with binding of fluorochromes to DNA, leading to flawed data. Tannic acid, a common phenolic compound, may be responsible for some of the stoichiometric errors, especially in woody plants. In this study, the effect of tannic acid on estimation of nuclear DNA content was evaluated in Pisum sativum and Zea mays, which were chosen as model species. - Methods Nuclear suspensions were prepared from P. sativum leaf tissue using four different lysis buffers (Galbraith’s, LB01, Otto’s and Tris.MgCl2).   • Průtoková cytometrie je metoda využívaná k určování ploidie a velikosti jaderného genomu. Pro zjišťování obsahu jaderné DNA jsou jádra v suspenzi barvena DNA-specifickým fluorochromem a fluorescence je kvantifikována. V této studii byl určován efekt kyseliny taninové při hodnocení obsahu jaderné DNA u Pisum sativum a Zea mays, které byly vybrány jako modelové druhy. Jaderná suspenze byla připravena z listové tkáně P. sativum s použitím čtyř různých lyzovacích pufrů (Galbraith’s, LB01, Otto’s and Tris.MgCl2). Suspenze byla ošetřena 13 koncentracemi kyseliny taninové (TA), od 0.25 do 3.50 mg mL-1. Po obarvení propidiem byly vzorky analyzovány pomocí FCM. TA ovlivnila fluorescenci PI a rozptylové vlastnosti světla jader rostlin, bez ohledu na použitý izolační pufr. Nejméně výrazný efekt TA byl pozorován u pufru Tris.MgCl2. Největší vliv na vzorky měly pufry Galbraith’s a LB01. Nově popsaný ´tannic acid effect´ se projevil ihned po ovlivnění. S výjimkou pufru Otto, jádra P. sativum a Z. mays byla ovli... (cs)
