Description
| - Dokázali jsme, že nadprodukce endogenních cytokininů způsobila stresové reakce v nekořenícím Pssu-ipt transgenním tabáku (Nicotiana tabacum L.), který je pěstován in vitro. Dokazuje to nadprodukce fenolických látek, syntéza pathogensis related proteinů a zvýšení aktivity peroxidas. Imunolokalizace zeatinu a PR-1b proteinu na kryořezech z listů prokázala jejich akumulaci ve všech buňkách transgenního tabáku ve srovnání s kontrolními netransgenními rostlinami. Intensivní modrá autofluorescence fenolických látek indukovaná UV v řezech listových žilek ukázala zvýšený obsah fenolických sloučenin v transgenním tabáku ve srovnání s kontrolou, nicméně nebyly zjištěny žádné významné rozdíly v celkovém obsahu ligninu v listech mezi oběma typy. Transgenní rostliny vykazovaly vyšší aktivity peroxidas jak rozpustné tak ionicky vázané frakce ve srovnání s kontrolou. - We present evidence that overproduction of endogenous cytokinins caused stress response in non-rooting Pssu-ipt transgenic tobacco (Nicotiana tabacum L.) grown in vitro. It was demonstrated by overaccumulation of phenolic compounds, synthesis of pathogenesis related proteins, and increase in peroxidase activities. Immunolocalization of zeatin and also PR-1b protein on leaf cryosections proved their accumulation in all mesophyll cells of transgenic tobacco contrary to control non-transgenic plants. Intensive blue autofluorescence of phenolic compounds induced by UV in cross-sections of leaf midrib showed enhanced contents of phenolics in transgenic tobacco compared with controls, nevertheless, no significant difference between both plant types was found in leaf total lignin content. Transgenic plantlets exhibited higher peroxidase activities of both soluble and ionically bound fractions compared with controls.
