Description
  • The pollen embryogenesis in Solanum nigrum L. (2n=6x=72) was successfully induced in all experimental and treated variants. The significant stimulation of androgenesis (numbered according to frequency of androgenic responsive anthers) was observed after 10 mM and 20 mM ethylmethanesulfonate (EMS), 10 mM and 20 mM sodium azide (NaN3) and 0.2 mM N-nitroso-N-methylurea (MNU) treatment. The frequency of androgenesis on medium with sucrose was significantly higher than on medium with maltose. The androgenic regenerants were obtained from anthers collected from donor plants where survival of mutagenic treatment was less than 50 %. We obtained green haploid (3x), aneuploid (to 8x) and dihaploid (6x) plants. We explain the high frequency of aneuploids among androgenic plants by irregularities in cell divisions in microsporial calluses.   • Pylová embryogeneze u druhu Solanum nigrum L. (2n=6x=72) byla úspěšně indukována u všech experimentálních variant. Významná stimulace androgeneze (vzhledem k frekvenci androgenně responzivních prašníků) byla zaznamenána po působení 10 mM a 20 mM ethylmethanesulfonátu (EMS), 10 mM a 20 mM azidu sodného (NaN3) a 0.2 mM N-nitroso-N-methylmočoviny (MNU). Frekvence androgenesis na médiu se sacharózou byla významně vyšší než na médiu s maltózou. Androgenní regeneranty byl odvozeny z prašníků rostlin, kde mutační působení přežilo méně než 50% rostlin. Zaznamenali jsme regeneraci zelených haploidních (3x), aneuploidních ( 8x) a dihaploidních (6x) rostlin. Vysoká frekvence aneuploidů byla zbůsobena asi poruchami jaderného dělení mikrosporiálních kalusů. Pozitivní účinek mutačního ovlivnění na indukci androgeneze může být vysvětlena zvýšením genetické různorodosti (pomocí bodových mutací) mezi buňkami uvnitř prašníku. (cs)
