Description
| - Pro mikrospory tabáku (Nicotiana tabacum L.) v období mitózy je charakteristický výskyt dominantních glykoproteinů 92 a 98 kDa (GP92a GP98). GP92 je rozpustná bílkovina, zatímco GP98 je vázán v nerozpustné frakci. Oba glykoproteiny byly izolovány afinitní chromatografií a pomocí SDS-PAGE a analyzovány pomocí MS. Sekvence peptidů byly stanoveny pomocí ?-HPLC/nano-ESI-MS/MS. GP92 je homologický k ?-galaktozidáze (EC 3.2.2.23) a GP98 k ?-xylozidáze (EC 3.2.1.37) Arabidopsis thaliana (L.) Heynh. Vývojové změny v aktivitě obou enzymů v extraktech mikrospor a pylu tabáku odpovídaly maximálnímu výskytu GP92 a GP98 v období mitózy mikrospor. Tyto dva glykoproteiny jsou první identifikované enzymy charakteristické pro mitotické mikrospory. Transkriptomické údaje týkající se exprese pěti genů pro ?-galaktozidázu a tří pro ?-xylozidázu v pylu Arabidopsis byly ověřeny RT-PCR na RNA získané z různých stádií vývoje pylu Arabidopsis a z různých částí sporofytu. - Tobacco (Nicotiana tabacum L.) microspores at the time of mitosis are characterized by the abundant occurrence of 92- and 98-kDa glycoproteins (GP92 and GP98). GP92 is a soluble protein while GP98 is bound to the insoluble microspore fraction. Both glycoproteins were isolated by affinity chromatography and SDS– PAGE and analysed by MS. Peptide sequences were determined by l-HPLC/nano-ESI–MS/MS (electrospray ionization tandem MS). GP92 displayed homology to b-galactosidase (EC 3.2.1.23) and GP98 to b-xylosidase (EC 3.2.1.37) from Arabidopsis thaliana (L.) Heynh. The activities of the two enzymes in microspore and pollen extracts of tobacco exhibited similar developmental changes to the occurrence of GP92 and GP98, with a maximum around microspore mitosis.
