  • Cytoskeletální bílkoviny vytvářejí dynamické polymery, které se uplatňují v mnoha procesech, v buněčném dělení, signální transdukci a v determinaci tvaru a polarity buňky. Distribuce a dynamika mikrotubulů (MTs) a aktinových mikrofilament (AFs) jsou mimo jiné determinovány lokalizací jejich nukleačních míst. Zatímco víme, že místo pro nukleaci rostlinných mitrotubulů je pod plasmatickou membránou a během interfáze na jaderném obalu, tak informace o nukleačních místech AFs jsou zcela nedostatečná. Tato práce využívá nízkou teplotu (0°C) k depolymeraci AFs a MTs u interfázních BY-2 buněk tabáku (Nicotiana tabacum L.). Rozsah depolymerace cytoskeletálních filament během chladového působení a subcelulární distribuce nukleačních míst během následného zotavení v 25°C bylo hodnoceno fluorescenční mikroskopií. Výsledky ukazují, že AFs rychle repolymerovaly z míst lokalizovaných v kortikální oblasti a na jaderném obalu, tedy podobně jako MTs. Na rozdíl od MTs, před kompletní rekostitucí A... (cs)

The distribution and dynamics of microtubules (MTs) and actin filaments (AFs) are determined, among other factors, by the location of their nucleation sites. Whereas the sites of microtubule nucleation in plants are known to be located under the plasma membrane and on the nuclear envelope during interphase, there is a striking lack of information about nucleation sites of AFs.
