  • Kombinace bioinformatických a experimentálních metodických přístupů byla užita k popsání a funkční charakterizaci nové rodiny kationtových přenašečů (CHX) u rostlin za použití modelového systému Arabidopsis. 28 CHX genů kóduje bílkoviny o Mw 85-95 kDa, které sestávají z N-koncové domény s 10-12 transmembránovými úseky (cca 440 aminokyselin) a hydrofilní domény (cca 360 aminokyselin) na C-konci, u které se předpokládá regulační úloha. Hydrofóbní, ale nikoli hydrofilní, doména rostlinných CHX je značně podobná monovalentním kation/proton antiporterům-2 (CPA2), zejména proteinům KHA1 (S. cerevisiae) a NhaS4 (Synechocystis). Poznatky o eukaryotických a prokaryotických CPA2 ukazují, že tyto bílkoviny fungují v několika transportních modech včetně K+/H+ (KHA1), NA+/H+-K+ (GerN) antiport a iontový kanál zprostředkovaný ligandem (KefC). Expresní profily genů AtCHX byle studovány pomocí RT-PCR, in situ expresí B-glucuronidasy řízenou promotory AtCHX genů a pomocí hybridisace Affymetrix ATH1 ... (cs)
  • A combined bioinformatic and experimental approach is being used to uncover the functions of a novel family of cation/H+ exchanger (CHX) genes in plants using Arabidopsis as a model. The predicted protein (85ů95 kD) of 28 AtCHX genes after revision consists of an amino-terminal domain with 10 to 12 transmembrane spans (approximately 440 residues) and a hydrophilic domain of approximately 360 residues at the carboxyl end, which is proposed to have regulatory roles. The hydrophobic, but not the hydrophilic, domain of plant CHX is remarkably similar to monovalent cation/proton antiporter-2 (CPA2) proteins, especially yeast (Saccharomyces cerevisiae) KHA1 and Synechocystis NhaS4. Reports of characterized fungal and prokaryotic CPA2 indicate that they have various transport modes, including K+/H+ (KHA1), Na+/H+-K+ (GerN) antiport, and ligand-gated ion channel (KefC).
