  • Prezentovaný článek popisuje stanovení aktivity tyroxin-hydroxylázy,která je jedním z enzymů v biosyntetické dráze linkomycinu. Metoda obsahuje vývoj HPLC metody, s isokratickou elucí a a fluorescenční detekcí. Při validaci této metody byl určen lineární rozsah pro DOPU 0.125–25 mmol/L s nejnižším limitem detekce LLOQ 0.125 mmol/L, RSD 7.2 percent a přesností 101.7 percent mmol/L a lineární rozsah pro tyroxin 15.625 mmol/L až 500 mmol/L s LLOQ 15.625 mmol/L, RSD 1.1 percent, a přesností 98.1 percent. Byla stanovena výtěžnost pro Dopa 100.66 percent s RSD 61472;0.89 percent a pro Tyr 94.76 percent s RSD 61472;0.94 percent. Byla stanovena přesnost a správnost metody v testu inter- and intra-day assay, která nepřekročila 10 procent. Dále byla určena stabilita vzorků reakční směsi při pokojové a 8722;20 stupňů C teplotě. Pomocí této metody bylo optimalizováno složení směsi enzymatické reakce a dále byly zjištěny podmínky pro zabránění vzdušné (neenzymatické) oxidace (cs)
  • An assay of L-tyrosine (Tyr) hydroxylating activity operating in lincomycin biosynthesis is described. The assay development consisted of HPLC procedure development, assessing the effect of reaction mixture components on non-enzymatic. Dopa and Tyr oxidation, and sample stability evaluation. The HPLC procedure with isocratic elution and fluorescence detection was developed and validated. The method showed a wide linear range of Dopa determination of 0.125–25 mmol/L with lower limit of quantification (LLOQ) of 0.125 mmol/L, RSD of 7.2 percent and accuracy of 101.7 percent. The studied linear range of Tyr was 15.625 mmol/L to 500 mmol/L with LLOQ of 15.625 mmol/L, RSD of 1.1percent, and accuracy of 98.1percent. Recoveries for Dopa and Tyr were 100.66 percent RSD 61472;0.89 percent and 94.76 percent RSD 61472;0.94 percent, respectively
