  • A novel alcohol oxidase (AOX) has been purified from mycelia of the wood degrading basidiomycete Gloeophyllum trabeum and characterized as a homooctameric nonglycosylated FAD flavoprotein with native and subunit molecular masses of 628 and 72.4 kDa. The isolated AOX cDNA coded for a polypeptide of 651 amino acids displaying 51–53% identity with other fungal AOX amino acid sequences. Using laboratory cultures and polyclonal antibodies, TEM immunogold labeling localized the enzyme in the hyphal periplasmic space and wall, and on extracellular tripartite membranes and slime, while there was no labeling of hyphal peroxisomes. AOX was further shown associated with membraneous/slime structures secreted by hyphae in wood fibre lumina and within secondary cell walls of degraded wood fibres

  • Nová alkoholoxidasa (AOX) purifikovaná z mycelia dřevokazného basidiomycetu Gloeophyllum trabeum byla charakterizována jako homooktamerický neglykosylovaný FAD flavoprotein o nativní a subjednotkové molekulární hmotnosti 628 a 72,4 kDa. Isolovaná AOX cDNA kódovala polypeptid o 651 aminokyselinách, vykazující 51–53% identitu s jinými publikovanými aminokyselinovými sekvencemi houbových AOX. Užitím polyklonálních protilátek a transmisní elektronové mikroskopie (imunoznačení zlatem) byla u laboratorních kultur AOX lokalizována v periplasmickém prostoru a buněčné stěně houbových hyf, na extracelulárních tripartitních membránách a v produkovaném slizu, zatímco v peroxisomech nebylo prokázáno žádné značení. AOX byla dále asociována s membránovými strukturami a slizem sekretovaným hyfami do dřevních vláken a uvnitř sekundární buněčné stěny degradovaného dřeva (cs)
