  • In this study, we performed proteomic analysis of porcine oocytes during in vitro maturation. Comparison of oocytes at the initial and final stages of meiotic division characterized candidate proteins that were differentially synthesized during in vitro maturation. While the biosynthesis of many of these proteins was significantly decreased, we found four proteins with increase in biosynthetic rate, which are supposed to play essential role in meiosis. Among them, the ubiquitin C-terminal hydrolase L1 (UCH-L1) was identified by mass spectrometry. To study regulatory role of UCH-L1 in the process of meiosis in pig model we used specific inhibitor of this enzyme, marked C30, belonging to the class of isatin O-acyl oximes. When germinal vesicle stage cummulus-enclosed oocytes were treated with C30 germinal vesicle breakdown was inhibited after 28 hours of culture and most of the oocytes were arrested at the first meiosis after 44 hours.   • V této práci byla provedena proteomická analýza prasečích oocytů zraných in vitro. Porovnáním oocytů na začátku a na konci meiotického dělení byly zjištěny významné proteiny, jejichž syntéza de novo byla rozdílná během zrání prasečích oocytů. Ačkoli biosyntéza mnoha z těchto proteinů byla výrazně snížena, byly nalezeny čtyři proteiny se zvýšenou mírou syntézy, o nichž se předpokládá, že mohou mít hlavní funkci během meiosy. Mezi nimi byla pomocí hmotnostní spektrometrie identifikována ubikvitin C- terminální hydloláza L1 (UCH-L1). Pro studium regulační funkce UCH-L1 v meiose prasečích oocytů byl použit specifický inhibitor tohoto enzymu, označený C30, který patří do skupiny isatin O-acyl oximů. Pokud byly oocyty ve fázi zárodečného váčku ošetřeny C30 inhibitorem, po 28 hodinách kultivace byl inhibován rozpad jaderné membrány, a dále oocyty kultivované v přítomnosti inhibitoru po dobu 44 hodin byly zastaveny ve fázi metafáze I.
