  • Equol je fyziologicky významný metabolit vznikající působením dosud neidentifikovaných střevních bakterií na daidzein a formononetin ? isoflavony vyskytující se v soje a dalších bobovitých rostlinách. Vykazuje estrogenní aktivitu, která je dokonce silnější než u jeho prekursorů. Přeměna daidzeinu na equol probíhá pouze u 30-50% lidské populace. V této studii jsme vyvinuli metody nepřímé kompetitivní ELISA pro stanovení equolu v biologickém materiálu (krev, moč). Jako imunogeny byly použity konjugáty equolu s hovězím sérovým albuminem vázané na nosič prostřednictvím karboxymethyletherových můstků v polohách C7 a C4?. Konjugáty stejných haptenů s ovalbuminem byly použity k imobilizaci. Jednotlivé ELISA metody byly optimalizovány testováním různých koncentrací protilátek a potahovacích antigenů, upřesněním reakčního času a využitím blokovacích činidel. Křížové reakce jednotlivých imunoanalytických metod s dalšími isoflavony jsou v přijatelném rozmezí (do 0.6%) a ovlinění molekulami s jinou strukturou j (cs)

The ability of individual subjects to convert daidzein to equol is highly variable. It has been assumed that equol may have higher biological activity than its precursor. Here we present the development of a competitive enzyme-linked immunosorbent assay (ELISA) method for the determination of equol in human body fluids. Isoflavone conjugates coupled to bovine serum albumin at positions 7-O- and 4´-O- via a carboxymethyl spacer were used as immunogens.
