  • Testy, založené na metodách molekulární biologie, se používají k identifikaci termofilních a mezofilních bacilů. Pět termofilních izolátů rodu Bacillus a tři mezofilní izoláty rodu Bacillus bylo podrobeno genotypizaci s využitím náhodně pomnožené polymorfické DNA (RAPD), s využitím amplifikace 16S-23S r RNA intergenového transkripčního spaceru (ITS-PCR), s využitím polymorfismu délky restrikčních fragmentů (RFLP) 16S r RNA a fragmentů ITS-PCR (ITS-PCR RFLP). Všechny uvedené metody byly optimalizovány tak, aby byly získány elektroforetogramy, z nichž bylo možné podle specifického vzoru rozlišit mezi jednotlivými druhy. RAPD protokol byl optimalizován s použitím dvou různých primerů, které oba se ukázaly jako vhodné pro RAPD fingerprinting. Po amplifikaci 16S-23S r RNA ITS bylo získáno po elektroforéze několik bandů, což vypovídá o několika kopiích ITS rozdílné délky u těchto bakterií. Vzory ITS bandů bylo snazší vyhodnotit než vzory RAPD, a proto se pro praxi ukázala jako vhodnější metoda ITS-PCR. V (cs)

A genotypic study using randomly amplified polymorphic DNA analysis (RAPD), amplification of the 16S-23S rRNA intergenic transcribed spacer (ITS-PCR), restriction fragment length polymorphism (RFLP) of 16S rRNA and restriction fragment length polymorphism of the ITS-PCR (ITS-PCR RFLP) was carried out with 5 thermophilic and 3 mesophilic Bacillus isolates. All above mentioned techniques have been optimized to be able to reach special electrophoresis pattern and discriminate among different species. RAPD protocol was optimized by using two different primers which both have been found to be effective for the RAPD fingerprinting of assessed bacilli. Amplification of the 16S-23S rRNA ITS produced multiple bands in gel electrophoresis, implying multiple copies of variable length in the ITS of these bacteria.
