| - During our work, flow cytometry was used for a study of the influence of cultivation conditions on a physiological state of the recombinant Pichia pastoris strain producing ?-galactosidase. The fluorescent dyes propidium iodide (PI) and bis-(1,3-dibutylbarburic acid) trimethine oxonol (BOX) were used for the determination of the structural integrity and membrane potential of Pichia pastoris cells, respectively. The optimized staining methods were employed in a study of a stress-induced physiological response to a change of substrate from glycerol to methanol in a glycerol steady state growing culture of Pichia pastoris yeast. During the first 5 hours of the transitional state an accumulation of methanol in the culture broth was accompanied by a decreasing concentration of biomass and an increasing amount of cells stained with PI and BOX. After the adaptation phase the amount of cells stained with PI and BOX reached steady levels of 2 % and 5 %, respectively.

- Během naší práce byl pomocí průtokové cytometrie studován vliv indukovaného stresového stavu na fyziologciký stav Pichia pastoris rekombinantního kmene produkujícího ß-galaktozidázu. Pro stanovení stukturální integrity cytoplasmatické membrány a membránového potenciálu byla použita fluorescenční barviva propidium jodid (PI) a bis-oxonol (BOX). Optimalizované metody byly využity ve studiu fyziologické odezvy kvasinky Pichia pastoris rostoucí v ustáleném stavu v chemostatu na indukovanou stresovou situaci vyvolanou změnou substrátu z glycerolu na metanol. Během prvních 5 hodin indukovaného stresu došlo k akumulaci metanolu v médiu, snížení koncentrace buněčné biomasy a zvýšení koncentrace buněk obarvených PI i BOX. Po fázi adaptace na nový substrát došlo opět k poklesu buněk barvených PI a BOX a jejich množství se ustálilo na hodnotách 2 % a 5 %. (cs)
