  • Tato práce je zaměřena na určení možností metody PCR (polymerásová řetězová reakce) k stanovování rodu Lactobacillus. PCR byla srovnávána s několika jinými metodami molekulární biologie, zvláště s metodou FISH (fluorescenční hybridizace in situ). Ze sbírky bylo vybráno 13 kmenů. Jejich identity byly potvrzeny testy API CH50. Zatímco biochemické vlastnosti všech vybraných kmenů laktobacilů jsou známy, kompletní genomová DNA sekvence je známa pouze u několika z nich. Pro jejich identifikaci bylo použito několik komerčně dostupných kitů, založených na PCR. Vzhledem ke znalosti sekvencí dobře uchovaných oblastí různých laktobacilových gDNA jsme schopni navrhnout primery pro tuto oblast. Tyto primery byly použity pro nespecifické PCR reakce s nižší teplotou annealingu. Reakce byla použita k určení sekvencí tohoto kmene, s neznámou sekvencí gDNA. Takto amplifikované DNA fragmenty byly klonovány do pCR #4 vektoru pomocí TOPO#TA kitu a poté sekvenovány. Přesná znalost sekvence nám dovolila navrhnout specifick (cs)
  • This work was focused on determination of possibilities of method PCR (polymerase chain reaction) to detect Lactobacillus. PCR was compared to several other methods of molecular biology, especially to FISH (fluorescence in situ hybridization). 13 Lactobacillus genera from collection were chosen. Their identities were confirmed by API CH50 tests. While biochemical features of all chosen Lactobacillus genera are known, the complete genomic DNA sequence is known only for some of them. Several commercially available kits based on PCR were used for their identification. Due to knowledge of a sequence of well-conserved region of different Lactobacillus gDNAs we could design primers for this region. These primers were used for non-specific PCR reaction with lower annealing temperature. The reaction was used for detection of the sequences from the genera with not known sequence of gDNA.
