  • Rhodobacter capsulatus belongs to group of purple nonsulfur bacteria. During the sequencing project Rhodobacter capsulatus SB1003 complete nucleotide sequence of its 3.7 Mbp chromosome and 133 kbp plasmid was determined. Three different clusters of genesencoding type I (RcpSBI, RcpSBII) and type III (RcpSBIII) DNA restriction and modification systems were found. Here we describe genetic and biochemical analysis of methyltransferase (MTase) RcpSBI. It comprises three subunits: one DNA sequence specifici ty subunit (HsdS) and two DNA modification subunits (HsdM). Expression vectors carrying hsdS and hsdM genes encoding MTase activity of RcpSBI were constructed. Both genes were expressed together in E. coli and complex of 44.2 kDa HsdS and 54.3 kDa HsdM was isolated by using affinity chromatography. The isoelectric point was determined to be 5.9.
