| - Nitric oxide (NO) is one of the free radicals that are implicated in a variety of intracellular signalling mechanisms. NO is synthesized from L-arginine by three different nitric oxide synthase (NOS) isoforms: neuronal NOS (nNOS), endothelial NOS (eNOS) and inducible NOS (iNOS). It has been suggested that NO-NOS has important physiological roles in a variety of reproductive processes such as follicular development, ovulation, and spermatogenesis. In the present study we investigated the role of NO-dependent events in meiotic resumption in pig oocytes. The mechanisms that promote the resumption of meiosis in mammalian oocytes remain poorly understood. INOS specific inhibitor AG significantly blocked the meiotic resumption of the oocytes in a dose-dependent manner. The addition of L-NAME to the culture medium had no significant effect on meiotic resumption and GVBD. Our results suggest that the iNOS-derived NO pathway plays important roles in pig oocyte meiotic maturation, especially in germinal vesicl
