Description
  • The EBC method 7.7, currently used for determination of bitter acids in hop products, is a time-consuming and laborious extraction technique. In this contribution, our aim was to propose a new extraction method based on Pressurized Fluid Extraction (PFE). Compared to conventional extractions, PFE offers a number of important benefits. PFE on OnePSE automated extractor was used for extraction of alpha- and beta-acids from hops and hop products. The parameters influencing the extraction efficiency were studied as well as the influence of the sample preparation method. The quantitative determination of alpha- and beta-acids in extracts was accomplished using an HPLC equipped with diode array detector. The experimental results were compared with those obtained by the standard EBC method 7.7.

  • V současnosti užívaná metoda stanovení hořkých kyselin chmele a chmelových produktů EBC 7.7 je časově nároční a pracná extrakční technika. Cílem naší práce bylo navrhnout novou extrakční techniku založenou na tlakové fluidní extrakci (PFE). V porovnání s konvenčními extrakcemi nabízí PFE řadu významných výhod. PFE na automatickém extraktoru OnePSE byla použita pro extrakci alfa- i beta-kyselin chmele a chmelových produktů. Byly studovány parametry ovlivňující výtěžnost extrakce i vliv metody přípravy vzorků. Jako koncovka stanovení alfa- i beta-kyselin byla použita HPLC s detekcí DAD. Bylo provedeno porovnání výsledků s výsledky získanými metodou EBC 7.7. (cs)
