  • V současné době metoda EBC 7.7 používaná pro stanovení hořkých kyselin ve chmelových produktech je pracná a časově náročná metoda. V této práci bylo naší snahou navrhnout novou extrakční metodu založenou na tlakové extrakci kapalin (PFE). PFE nabízí mnoho významných předností. K PFE extrakci hořkých alfa- a beta-kyselin z chmele a chmelových preparátů byl využit automatizovaný extraktor OnePSE. Byly studovány parametry ovlivňující výtěžnost extrakce stejně jako způsob přípravy vzorků. Kvantitativní stanovení obsahu hořkých alfa- a beta-kyselin v extraktech bylo uskutečněno pomocí vysokoúčinné kapalinové chromatografie a detekce pomocí detektoru s diodovým polem. Získané výsledky byly porovnány s výsledky získanými standardní metodou EBC 7.7. PFE metodu lze pokládat za perspektivní metodu a možnou alternativu k metodě EBC 7.7. (cs)
  • The EBC method 7.7, currently used for determination of bitter acids in hop products, is a time-consuming and laborious extraction technique. In this contribution, our aim was to propose a new extraction method based on Pressurized Fluid Extraction (PFE). Compared to conventional extractions, PFE offers a number of important benefits. PFE on OnePSE automated extractor was used for extraction of alpha- and beta-acids from hops and hop products. The parameters influencing the extraction efficiency were studied as well as the influence of the sample preparation method. The quantitative determination of alpha- and beta-acids in extracts was accomplished using an HPLC equipped with diode array detector. The experimental results were compared with those obtained by the standard EBC method 7.7. PFE method presents a promising alternative to the EBC method 7.7.activities.
