  • Byla vypracována chromatografická metoda pro separaci isoflavonoidů (daidzein, genistein, formononetin a biochanin A) a současně některých flavonoidů (rutin, myricetin, quercetin a apigenin) na reversní stacionární fázi ZORBAX-SB C18. Byly vypočítány limity detekce a meze stanovení a posouzeny možnosti přesnosti a správnosti stanovení také s ohledem na způsob přípravy analyzovaných vzorků (extrakce a následné ředění mobilní fází). Získané výsledky ukázaly, že chmel může být bohatým zdrojem rutinu, quercetinu, 4-hydroxykumarinu, myricetinu a naringinu. Tyto látky byly též nalezeny v pivu, některé však v koncentracích blížících se mezi detekce. Fytoestrogeny byly detekovány ve chmelu i pivu, koncentrace byly též blízko limitů detekce. Ředění vzorků mobilní fází je optimální v případě vysokých obsahů některých flavonoidů. Ale pro nízké koncentrace isoflavonouidů je nezbytné najít jiný způsob přípravy vzorků, např. SPE. (cs)
  • The HPLC method for separation of some isoflavonoids (daidzein, genistein, formononetin and biochanin A) from beer or hops in addition to some flavonoids (rutin, naringin, myricetin, quercetin and apigenin) was elaborated using phase ZORBAX-SB C18. The detection limits and limits of estimation were calculated. The possibilities of accuracy of determination of analysed compounds were evaluated by involving of the described manners of samples preparation. The obtained results showed that hops could be a rich source of rutin, quercetin, 4-hydroxycoumarin, myricetin and naringin. Consequently these compounds were found also in beer, but measured concentrations were often near to limits of detection. Phytoestrogens were detected in hops and also in beer, but their quantification was also near to limits detection. The dilution of samples is optimal in case of rich contents of some flavonoids. But for low concentration of isoflavonoids it is necessary to find another way of sample preparation, e.g. SPE.
