  • Biogenní aminy pocházejí ze surovin a fermentačních fází výroby piva. Tyto látky jsou toxické :jejich obsah by neměl překročit hygienické limity, a proto jsou pečlivě kontrolovány. Zejména pro histamin, tyramin, tryptamin, putrescin a kadaverin byla vypracován postup pro rutinní analýzu metodou HPLC s fluorescenční detekcí (ex = 330 nm, em = 440 nm) a automatickou předkolonovou derivatizací s ortho-phthaldehydem v prostředí borátového pufru. Aminy byly izolovány ze vzorků piva na nové stacionární fázi Discovery DSC-MCAX a po čistícím kroku byly vymyty alkalizovaným methanolem (1% NaOH). 1,7-diaminoheptan byl použit jako vnitřní standard pro kontrolu derivatizačního procesu. Jednoduchá příprava vzorku a automatický proces HPLC systému umožňují rychlé stanovení biogenních aminů v pivu. (cs)
  • The biogenic amines in beer originate from the raw materials and the fermentation phases of beer production. These compounds are toxic: their contents should not exceed hygienic limits and are therefore carefully controlled. Predominantly histamine, tyramine, tryptamine, putrescine and cadaverine were routinely analysed by HPLC with fluorescence detection (ex = 330 nm, em = 440 nm) and an automatic precolunm derivatization with ortho-phthaldehyde in the presence of a borate buffer. The amines were isolated from beer samples in the new stationary phase Discovery DSC-MCAX and after a clean–up procedure were washed with alkalized methanol (1% NaOH). 1,7-diaminoheptan was used as an internal standard to check the derivatization procedure. Easy sample preparation and the automatic operating HPLC system enable a speedy determination of biogenic amines in beer.
