  • In mice, the Bcg locus of the chromosome 1, also known as Ity or Lsh , has been implicated in natural resistance or susceptibility to infection with unrelated intracellular pathogens including Mycobacterium bovis BCG and several other mycobacterial species [1, 2], Salmonella typhimurium [3], Leishmania donovani [4], and Francisella tularensis [5]. The genetic control is manifested in the initial phase of infection when it is characterized by a rapid microbial replication in susceptible mice, as opposed to controlled proliferation in resistant animals. Macrophage is the cell type responsible for the phenotypic expression of genetic difference at Bcg [6]. Using positional cloning, a candidate for Bcg was identified and designated Nramp1 (natural resistance-associated macrophage protein 1) [7]. Gene knockout and transgenesis provided final verification that the Nramp1 gene is implicated in resistance to taxonomically and antigenically unrelated microorganisms controlled by the Bcg/Ity/L
