Description
| - Patatin is a glycoprotein and its fuction is as a storage protein but has been demonstrated some enzyme activities - lipd acyl hydrolase (LAH) activity is the most important. LAH activity mayplay a role in the plant defense mechanisms but physiological roles is not complete understood. Patatin has been found in all examined potato cultivars, including the South American genotypes of groups Andigena, Phureja and Stenotomum. Potato tubers were donated by the Potato Research Institute Havlíčkův Brod, Czech Republic. The presented contribution is focused on lipid acyl hydrolase activities (LAH) of the major potato tuber protein patatin of selected genotypes from South America: four genotype Solanum andigenum, two genotypes Solanum goniocalyx, three genotypes Solanum phureja and five genotypes Solanum stenotomum and for comparison Solanum tuberosum cv. Desireé. LAH was determined qualitatively (on the gel) and quantitatively (by spectrophotometry). Detection of LAH on the polyacrylamide gel showed from 1 to 4 protein bands with the relative mobility 0.75-0.89. Quantification of LAH specific activitity resulted in the interval from 2.6 (cv. Desireé) to 23.2 (S. stenotomum) umol/(min mg). The procedure is based on a assay using p-nitrophenyl burytate as the enzyme in absorbance at 410 mm. Quantification of LAH activity was followed by determination of Michaelis - Menten kinetics parameters. The relative abundance of patatin was detected using automatic chip electrophoresis Experion and was ranged from 15.2 (cv.Desireé) to 40.6 (S.goniocalyx) %. In the selected set of genotypes was confirmed the variability LAH of the patatins. The results showed that four use LAH in practice can be non-traditional genotypes of potatoes a soure of proteins with higher levels of activity.
