  • Bylo prováděno srovnání kultivace rostlin na médiu MS a na médiu SH z hlediska výtěžnosti izolovaných protoplastů. Výtěžnost protoplastů izolovaných z rostlin kultivovaných na SH médiu byla lepší v případě, že kultivované rostliny byly pasážovány ve stadiu aktivního růstu. U kalusů protoplastového původu byla testována reakce na odstupňované koncentrace 1:5 IAA:ZT a odlišné poměry 1:4 až 1:25 IAA a ZT za účelem zkrácení doby potřebné k regeneraci rostlin na kalusech. Experimenty byly uskutečněny v tkáňových destičkách 4 x 6 jamek. Růstové regulátory byly aplikovány v MS médiu. Doba aplikace byla 90 dnů. Perspektivní kombinace byla 0,1 mg/1 IAA, 2 mg/l ZT. Doba potřebná pro zahájení regenerace rostlin na kalusech se zkrátila v průměru o 1 měsíc ve srovnání s předchozími zkušenostmi. Pozitivní efekt byl pozorován zejména na kalusech po fúzích protoplastů. Postupně zvyšovaná koncentrace růstových regulátorů v poměru 1:5 auxin:cytokinin nevedla k regeneraci rostlin. (cs)
  • Protoplast yield from plants cultivated on MS medium were compared with SH medium. Protoplast yield from plants cultivated on SH medium was unambiguously better in case when donor plants were maintained and subcultured on the medium in the stage of intensive growth.The protoplast-derived calli were tested for response to the graded contents of the plant hormones with ratio 1:5 IAA:ZT and to the contents with different ratios from 1:4 to 1:25 IAA and ZT with aim to short time needed for organogenesis. The experiments were performed in tissue-culture plates with 4 x 6 wells. The application period was 90 days. The perspective combination was 0.1 mg/1 IAA, 2 mg/l ZT. The necessary time for start of shoot regeneration was in average 1 month shorter as compared to earlier experience. The positive effect was observed especially on protoplast-derived calli after electrofusion. A gradually increased concentration of growth regulators with the ratio 1:5 auxin:cytokinin did not result in plant regeneration.
