  • Byla vybrána optimální metoda aklimatizace somatických hybridů rodu Solanum z in vitro do in vivo podmínek. (cs)
  • Many valuable resistance genes are carried by wild species of Solanum genus that are often non-crossable with cultivated potato. Somatic hybridization is an approach overcoming this problem. The somatic hybrids were developed through protoplast fusion by an electric field (Navrátilová et al. 2006), they were evaluated by flow-cytometry, RAPD technique and their in vitro morphology. Optimal method for the most effective transfer into in vivo conditions was investigated. A set of somatic hybrids of two combinations was planted into seedling pots (3x3 cm) containing sterilized soil and cultured in a laboratory with artificial illumination (4000 Lux) of 16 h light/8 h dark photoperiod at 21 °C. The second variant involved direct planting into the pots (8x8 cm). Plants planted in this way were partially protected by non-woven fleece and exposed to weather conditions. Rate of survival and growth increment was investigated in both variants.
