Description
  • Pro modelovou fúzi protoplastů byly vybráni zástupci rodu Solanum, kteří pozitivně reagovali v protoplastové kultuře a dobře regenerovali až do stádia rostlin. Cílem modelové fúze bylo ověřit vhodnost optimalizovaných parametrů elektrického pole, a tak získat somatické hybridy ve stádiu kompaktních rostlin v co nejkratší době. Je popsána metodika izolace protoplastů, fúze a kultivace až do dosažení regenerace rostlin, které byly analyzovány pomocí průtokové cytometrie a analýzy DNA metodou RAPD. Optimální velikost střídavého proudu pro zřetězení protoplastů byla 95 – 100 V/cm s individuální dobou působení (2 – 15 s) a stejnosměrného proudu pro vlastní fúzi byla 190 – 200 V/cm s trváním 80 µs. Celkový počet regenerovaných rostlin bylo 579, z toho 25 somatických hybridů. Je diskutována úspěšnost somatické hybridizace a její využití jako alternativního nástroje šlechtění u brambor. (cs)
  • The representatives of genus Solanum, which positively responded to protoplast culture and achieved the plant regeneration well, were chosen for model electrofusion of protoplasts. The aim of model fusion was to verify the suitability of optimised of electric field parameters and to obtain somatic hybrids as the compact plants in short time. It is described the procedure of protoplast isolation, fusion and culture to the plant regeneration. The optimal doses for successful fusions were 95 - 100 V/cm AC with individual operation time (2 - 15 s) followed by 190 - 200 V/cm DC for 80 µs. Cut plants were estimated by the flow cytometry and DNA analysis namely RAPD. Total number of regenerated plants was 579, RAPD revealed 25 somatic hybrids. The fruitfulness of somatic hybridisation and its utilisation as alternative breeding tool is discussed.
