Description
  • Protoplasts were isolated from in vitro cultured plants or calluses. Enzymatic mixture of 1% or 2% Cellulase Onozuka R-10 and 0,25% or 1% Macerozyme R-10 was used. Purified protoplasts were rinsed with pre-fusions solution and fusion solution. Viability of isolted protplasts used for electrofusions was more than 80%. Protoplast fusion was performed using apparatus ECM 2001 and in electroporation chamber for 20-30 ul with the 0,5 mm electrode distance. Following parameters were used for fusions: for protoplasts alignment - 5 V AC and 1 MHz frequency with individual duration of action, for fusion - 1 pulse 10 V DC of length 80 us. Subsequent culturing was done in medium SW11. Regeneration of cell walls was recorded and microcolonies (Solanum) were found in 15 days.

  • Protoplasty byly izolovány z rostlin nebo kalusů kultivovaných in vitro. Byla použita enzymatická směs 1% nebo 2% Cellulase Onozuka R-10 a 0,25% nebo 1% Macerozyme R-10. Purifikované protoplasty byly promyty v předfúzním roztoku a ve fúzním roztoku. Životnost izolovaných protoplastů použitých pro fúze byla více než 80%. Fúze protoplastů se uskutečnila pomocí přístroje ECM 2001 v elektroporační komůrce pro objem 20 - 30 ul se vzdáleností elektrod 0,5 mm. Pro fúze byly použity tyto parametry: k seřazení protoplastů - 5 V střídavého proudu při freikvenci 1 MHz s individuální dobou působení, pro fúzi - 1 puls 10 V střídavého proudu s dobou trvání 80 us. Následující kultivace po fúzi probíhala v médiu SW11. Byla zaznamenána regenerace buněčných stěn a mikrokolonie (Solanum) se objevily po 15 dnech. (cs)
