  • Popsali jsme mitochondriální genom a ultrastrukturu u Diplonema papillatum, příslušníka diplonemích bičíkovců, kteří jsou sesterskou skupinou kinetoplastid. Fluorescenční a elektronová mikroskopie prokázala přítomnost jediné mitochondrie v buňce a ultrastrukturu atypickou pro Euglenozoa. Navíc je DNA rovnoměrně v organele rozptýlená a není kompaktně sbalené v kinetoplastu. Molekulární analýzy prokázaly mnohočetné malé mitochondriální chromosomy příslušející ke dvěma třídám o velikosti 6 a 7 kbp. In vivo mají tyto molekuly monomerní kruhovou topologii a jsou relaxované. Je pozoruhodné, že gen pro cox1 (a pravděpodobně i další geny) jsou fragmentované s tím, že jednotlivé kousky jsou uložené na různých chromosomech. Tvorba souvislé mRNA tudíž vyžaduje trans-splicing, jehož mechanismus zatím není znám. (cs)
  • We describe the mitochondrial genome and the mitochondrial ultrastructure of Diplonema papillatum - a member of diplonemid flagellates, which are the sister group of kinetoplastids. DNA is evenly distributed throughout the mitochondrion rather than compacted into a kinetoplast body. Molecular studies distinguish numerous small mitochondrial chromosomes that fall into two size classes, 6 and 7 kbp. In vivo, these molecules are of monomeric circular topology and relaxed conformation. Remarkably, the cox1 gene (and probably other mitochondrial genes) is fragmented, with separate gene pieces encoded on different chromosomes. Generation of the contiguous cox1 mRNA requires trans-splicing, the precise mechanism of which remains to be determined. Taken together, the mitochondrial gene/genome structure of Diplonema is not only different from that of kinetoplastids but unique among eukaryotes as a whole.
