Description
| - Ukázali jsme, že jaderný receptor NHR-25, jehož ortology u drosofily a u savců jsou důležitými regulátory vývoje a pohlavní determinace, hraje kritickou roli v diferenciaci epidermis. Nedostatek NHR-25 zabraňuje epiteliálním kmenovým buňkám obnovovat jejich vzájemná spojení, která jsou dočasně přerušována asymetrickými děleními těchto buněk. Ztráta těchto mezibuněčných kontaktů vede k abnormální specifikaci osudu dceřinných buněk a k jejich nadpočetným dělením. Model C. elegans tak ukázal novou úlohu evolučně konservovaného jaderného receptoru na úrovni diferenciace jednotlivých buněk. (cs)
- We show that a nuclear receptor NHR-25, whose Drosophila and mammalian orthologs are important regulators of development and sex determination, plays a critical role in epidermal differentiation. Deficiency of NHR-25 prevents epithelial stem cells to renew their adherens junction contacts, which are temporarily lost following asymmetric divisions of these cells. The lack of these cell-cell contacts results in abnormal fate specification of the stem cell daughters and in their excessive division. The C. elegans model reveals a new role for the evolutionarily conserved nuclear receptor with an individual cell resolution.
