  • Nový, polyfázický přístup byl aplikován při studiu diversity sinic v letním fytoplanktonu českých přehrad v letech 2001 a 2002. Přístup spočívá v současném použití tradičních (mikroskopická analýza druhového složení a stanovení biomasy) a molekulárních metod (srovnání genetických sekvencí 16S rRNA získaných pomocí DGGE s klonovými knihovnami). Mikroskopickou analýzou 38 vzorků planktonu bylo nalezeno 15 rodů sinic, pomocí DGGE bylo však bylo odděleno 113 sekvencí. Identifikace dominantních druhů se shodovala u 32 vzorků. Značné rozdíly byly však zjištěny u nepatrných populací a u velmi malých druhů rodů Aphanothece, Merismopedia, Pseudanabaena, Romeria, Snowella and Synechococcus. Tyto rozdíly lze vysvětlit zvláštnostmi použitých metod (competitivní amplifikace přo PCR, obtížemi při vymývání DGGE pásků a velmi malými populacemi, které nemohly být v mikroskopu zaznamenány.)   • We used a polyphasic approach combining data from microscopic assessment of fresh biomass and from clone libraries and DGGE fingerprints based on 16S rRNA gene sequences to investigate the cyanobacterial diversity of Czech reservoirs during the summer in 2001 and 2002. Total of 15 genera were identified using the microscopic analysis in 38 samples analysed. We recovered 113 DGGE band sequences from the same samples. Identification of the major populations by the microscopic enumeration and the molecular results were generally congruent (for 32 samples out of 38). Differences concerned the presence of minor populations of minute genera. Aphanothece, Merismopedia, Pseudanabaena, Romeria, Snowella and Synechococcus. These differences can be explained by biases specific to each method (competitive amplification, difficulty to obtain sequences from DGGE bands, problems of microscopic observation of the small-sized genera).
