  • Soil microbial community of colliery spoil heaps during primary succession (succession plots: initial [0] - early [10] - mid [20] - late [42 years old], Sokolov, North-West Bohemia) was studied by using both phenotyping methods (phospholipid fatty acids - PLFA pattern, carbon utilisation patterns - Biolog System, dominant heterotrophic bacterial species - Sherlock MIDI System) and genotyping method (ARDRA - Amplified Ribosomal DNA Restriction Analysis). PLFA pattern was clearly different among individual succession stage as well as among the top (0-5 cm) and the mineral (10-15 cm) layer. The PLFA biomarkers indicated that the initial and the early stages were enriched by actinobacteria; the mid stage by Gram negative bacteria and fungi; the late stage was characteristic by the development of microeukaryota.   • Bylo studováno společenstvo půdních mikroorganismů na hnědouhelných výsypkách (sukcesní plochy: počáteční 0 – raná 10 – prostřední 20 – pozdní 42 let stará, lokalita Sokolov, Severozápadní Čechy) pomocí fenotypových metod (Sherlock MIDI System – screening dominantních heterotrofních druhů, PLFA – fosfolipidické mastné kyseliny, Biolog System - analýza využití C – substrátů) a genotypových (ARDRA – restrikční analýza amplifikované ribozomální DNA). PLFA profily se lišily mezi jednotlivými sukcesními stádii a zároveň mezi vrchní (0-5 cm) a minerální (10-15 cm) vrstvou. Počáteční a rané stádium byly bohaté na aktinobakterie, v prostředním převažovaly Gram negativní bakterie a houby; pozdní stádium bylo charakteristické vývojem microeukaryot. Funkční diverzita byla nejvýraznější mezi pozdní fází sukcese a mladšími stádii.
