  • Recently we studied 118 Borrelia isolates, cultured from samples collected from a variety of birds, rodents and ticks in 9 different localities of the southern region of the Unites States. A group of 16 isolates with unusual characteristics was found. The new methodology of multilocus sequence analysis (MLSA) that involved the study of rrf-rrl intergenic spacer region, rrs (16S RNA) gene, flagellin gene, ospA gene and p66 gene was used to clarify the taxonomic status of this new highly homogeneous group. All 13 recognized borrelia species were used as controls. The association of this species with Lyme disease group was confirmed by multiple results of MLSA. Unique RFLP pattern was discovered in the rrf-rrl intergenic spacer and flagellin gene. Unique signature nucleotides were allocated in the 16S rRNA. Phylogenetic analysis based on concatenated sequences of 5 loci showed all 16 isolates clustered together but separately from the other species of B. burgdorferi s.l. complex.

  • V poslední době jsme studovali 118 izolátů borelií kultivovaných ze vzorků sebraných od ptáků, hlodavců a klíšťat v 9 různých lokalitách na jihu Spojených Států. Byla identifikována skupina ze 16 izolátů s neobvyklými charakteristikami. Pro vyjasnění taxonomického statusu nové vysoce homogenní skupiny vzorků byla použita nová metoda multilocus sequence analysis (MLSA) která zahrnuje studium rrf-rrl intergenic spacer regionu, rrs (16S RNA) genu, genu pro flagelin, ospA a genu pro p66. Všech 13 uznaných druhů borelií bylo použito jako kontroly. Sounáležitost nově objevených vzorků do skupiny spirochét Lymské boreliózy byla potvrzena četnými výsledky MLSA. Jedinečný RFLP profil byl objeven v rrf-rrl intergenic spacer regionu a genu pro flagelin. Jedinečné signature nukleotidy byly určeny v 16S rRNA. Fylogenetická analýza všech 5 studovaných regionů ukázala, že všech 16 izolátů se seskupuje do jednoho klasteru, ale samostatně od ostatních druhů komplexu Borrelia burgdorferi sensu lato. (cs)
