  • Práce se zabývá měřením deposice Si u přírodních populací rozsivky Fragilaria crotonensis, které jsme prováděli na římovské nádrži v roce 2006 za pomocí fluorescenčního značení PDMPO. Rozsivky byly zastoupeny téměř výhradně druhem F. crotonensis, který tvořil výrazná podpovrchová maxima biomasy (PMB, hloubka 2-6,5m). Deposice Si byla osmkrát vyšší u hladiny než v hloubce PMB a také procento značených buněk bylo podstatně vyšší (50 % u hladiny, 20 % v hloubce PMB). Nízká rychlost depozici Si v hloubce PMB byla zapříčiněna nedostatkem světla, ačkoli naměřené hodnoty světelného záření se pohybovaly kolem 1% u hladiny, což je tradičně považováno za dostatečné pro fotosyntézu. U hladiny bylo ukládání Si během roku ovlivněno nedostatkem fosforu. Tato studie představuje první pokus o propojení PDMPO techniky s kvantitativním měřením fluorescence pomocí analýzy obrazu. Na základě našich výsledků, PDMPO představuje užitečný nástroj pro studium ekologie přírodních populací rozsivek. (cs)
  • In 2006, we measured Si deposition in the summer diatom assemblage at various depths in a eutrophic Římov Reservoir using PDMPO technique. Diatom assemblage was exclusively dominated by Fragilaria crotonensis, which formed pronounced subsurface maxima. Concentrations of the main nutrients were low over the whole season, however, at depth the nutrient availability was higher than at the surface. Fragilaria Si deposition rates were eight times higher at the surface than at depth. A half of the population was involved in Si deposition at the surface while only 20 % active cells were found at depth. At the surface, Si deposition was limited by phosphorus deficiency; the effect of dissolved Si was not statistically significant. Si deposition at depth was significantly constrained by low light availability despite the 1 % average light attenuation at depth, which is supposed sufficient for photosynthesis.
