  • Bránit předčasné metamorfose hmyzu je dlouho známou funkcí juvenilního hormonu (JH), avšak mechanismus účinku JH ani jeho receptor dosud neznáme. Nadějným kandidátem na roli receptoru JH je protein Methoprene-tolerant (Met), jehož ztráta u drosofily způsobuje resistenci k JH, ale nikoli předčasnou metamorfosu. S využitím modelu brouka Tribolium castaneum jsme ukázali, že umlčení homologního genu vede v souladu s anti-metamorfním účinkem JH k předčasnému kuklení a k dalším heterochronním znakům předčasné metamorfosy. Met tak v odpovědi na JH zprostředkuje správné načasování vstupu larvy do metamorfosy. Naše výsledky poprvé dokazují, že Met je nezbytný pro signalizaci JH během hmyzího vývoje, a podporují dosud zpochybňovanou funkci Met jako receptoru JH. (cs)

One promising candidate for the JH receptor role is the Methoprene-tolerant protein (Met), whose loss renders Drosophila resistant to JH but fails to cause precocious metamorphosis. By using the beetle model Tribolium castaneum, we show that silencing of the Met homolog leads to premature pupation and to other heterochronic phenotypes marking precocious metamorphosis, an effect consistent with the anti-metamorphic JH function. In response to JH, Met thus mediates proper timing of entry to metamorphosis. Our data for the first time demonstrate that Met is necessary for developmental JH signaling, and support the disputed role of Met in JH reception.
