  • TNF-alpha plays an important role in the initiation of inflammation. We identified a TNF-alpha-inhibitory activity in the saliva and salivary gland extract (SGE) from partially fed Ixodes ricinus females. Using mouse and human TNF-alpha-specific ELISA, we showed that saliva or SGE reduced the level of detectable cytokine. Both saliva and SGE inhibited the cytotoxic effect of TNF-alpha. Elimination of the TNF-alpha-inhibitory activity in SGE by trypsin digestion demonstrated that the anti-TNF-alpha factor is a protein. FPLC fractionation of SGE showed one peak of TNF-alpha-inhibitory activity corresponding to a protein with estimated molecular mass 23 kDa. The likely mechanism of the inhibitory effect is direct binding of the cytokine. The TNF-alpha-inhibitory molecule seems to play an important role in the anti-inflammatory effect of tick saliva at the tick feeding site, providing a gateway to the host for tick-borne pathogens.

  • Faktor nekrotizující nádory (TNF-alfa) je jedním z nejdůležitějších mediátorů zánětu. Ve slinách a v extraktu ze slinných žláz (SGE) částečně nasátých samic klíšťat Ixodes ricinus jsme identifikovali TNF-alfa inhibující aktivitu. Pomocí ELISA testu pro TNF-alfa jsme ukázali, že klíštěcí sliny nebo SGE výrazně snížily hladinu detekovatelného cytokinu. Sliny a SGE inhibovaly cytotoxický efekt TNF-alfa v biologickém testu. Pomocí eliminace TNF-alfa-inhibiční aktivity v SGE trypsinovou digescí jsme prokázali, že anti-TNF faktor je protein. Frakcionace SGE pomocí FPLC ukázala jeden vrchol TNF-alfa-inhibiční aktivity odpovídající proteinu o molekulové hmotnosti 23 kDa. Pravděpodobným mechanismem inhibičního efektu je přímá vazba cytokinu. Zdá se, že TNF-inhibující molekula hraje důležitou roli v protizánětlivém účinku klíštěcích slin v místě sání klíštěte a usnadňuje tak vstup patogenů přenášených klíšťaty do hostitele. (cs)
