  • An experiment designed to examine food preferences of heterotrophic nanoflagellates grazing on bacterioplankton was performed in the freshwater Římov reservoir. Water samples were size-fractionated to obtain <5um filtrate containing bacteria and HNF. To manipulate resource availability,<5um treatments were incubated in dialysis bags submerged in the barrels filled with the unfiltered reservoir water amended with either orthophosphate or glucose or combination of both. We employed r-RNA-targeted probes to assess HNF prey preferences by analyzing bacterial prey in HNF food vacuoles compared to available bacteria. Actinobacteria were avoided by HNF in all treatments. Cytophaga-Flavobacterium-Bacteroidetes bacteria were positively selected mainly in treatments in which bacteria were heavily grazed,the <5um treatments, but this trend was less pronounced towards the end of the study. The members of a small subcluster of Betaproteobacteria were mostly positively selected.

  • Experiment sledující potravní preference prvoků byl proveden na sladkovodní nádrži Římov. Vzorky vody byly velikostně frakcionovány, abychom dostali frakci menší než 0,5 mikrometrů obsahující bakterie a heterotrofní prvoky. Abychom mohli manipulovat dostupnost nutrientů, 5 mikrometrové frakce byly inkubovány v dialyzačních sáčcích ponořených v barelech naplněných nefiltrovanou vodou z nádrže, obohacenou o fosfát, glukózu, případně obojí. Byl aplikovány genetické próby přímo k určení potravních preferencí prvoků analýzou jejich potravních vakuol. Bakterie patřící mezi Actinobacteria byly negativně selektovány ve všech variantách. Bakterie patřící do skupiny Cytophaga-Flavobacterium-Bacteroidetes byly vesměs pozitivně selektovány, hlavně ve variantách se silným predačním tlakem vyvíjeným na bakterie. Tento trend se měnil s postupujícím pokusem. Podskupina bakterií patřících do skupiny Betaproteobacteria byla prvoky pozitivně přijímána. (cs)
