  • Trypsin-like activity, similar to acrosin, is present in sturgeon spermatozoa and can be a potential target for trypsin inhibitors. The objective of this work was to use a fluorescent soybean trypsin inhibitor (SBTI) conjugate with the Alexa Fluor? 488 dye for visualization of sturgeon acrosome. After incubation with SBTI-Alexa, a strong signal was observed both in the acrosome and midpiece or implantation fossa regions. We have also found that SBTI-Alexa staining can be combined with live/dead staining. Close examination of staining pattern revealed that SBTI-Alexa can either stain acrosome or whole sperm. Staining of whole sperm correlated with live/dead staining (r2 = 0.94, p < 0.01). However, in fresh semen most cells (93 - 97%) were not stained with SBTI-Alexa, probably due to intact acrosomes. Further studies should test if SBTI-Alexa can be applied to monitor the acrosome status during the acrosome reaction and cryopreservation.

  • Aktivita trypsinu, podobně jako akrosinu, je přítomna ve spermiích jeseterů a může být potencionálním cílem pro inhibitory trypsinu. Cíl této práce bylo užití Soybean Trypsin Inhibitoru (SBTI) spojeným s Alexa Fluor? 488 fluorescenční barvy (Alexa) pro vizualizaci akrosomu. Po inkubaci Alexou byl pozorovaný silný signál v akrosomu a středním oddílu či implantační jamce. Také jsme zjistili, že Alexa může být kombinována s live/dead staining. Bližším pozorováním modelového barvení jsme odhalili, že Alexa může značit jak akrosomu tak celou spermii. Barvení celých spermií korelovalo s live/dead barvením (r2 = 0.94, p < 0.01). Nicméně, u čerstvých vzorků většina spermií (93 - 97%) barvena Alexou nebyla, pravděpodobně kvůli neporušeným akrosomům. Další studie by měli testovat zda-li může být Alexa užita pro monitorování stavu akrosomu během akrosomové reakce a kryokonzervace. (cs)
