| - The main aims of the present work were to examine effects of potassium, calcium and immobilizing medium on motility of fresh and demembranated spermatozoa of paddlefish (Polyodon spathula).Evaluation of sperm motility parametersSperm motility was evaluated for the percentage of motile spermatozoa from video frames, velocity was measured as the distance covered by sperm heads during the corresponding to three video frames and frequency was measured using dark-field microscopy coupled with stroboscopic illumination.Short-term storage of paddlefish sperm in immobilizing mediumSeminal plasma of paddlefish has a low spermatozoa concentration (around 0.1-1x109 sperm), therefore it was necessary to concentrate cells by centrifugation (2000 g for 5 min). For examination, 100 #l of centrifuged seminal fluid (transparent part) was diluted with following immobilizing solution (IM): 900#l of seminal fluid, 800 #l of seminal fluid and 100 #l DMSO, 900#l of SM containing 5 mM KCl, 800 #l of SM containing 5 mM KCl
