  • Polymorfismus náhodných sekvencích (RAPD) byl použit k detekci intraspecifické diverzity u L. donovani komplexu. Bylo použito 52 primerů (deseti až jednadvaceti merů), které amplifikovaly náhodné sekvence u 15 kmenů příslušícím k devíti zymodemovým skupinám. Jako outgroup byly použity L. major a L. tropica. Bylo generováno 902 amplikonů, z nichž pouhých 4.3% bylo shodných pro všech 15 kmenů L. donovani komplexu. Byl zkonstruován dobře podpořený fylogenetický strom, který podpořil silnou korelaci mezi genetickým polymorfismem a geografickýn původem. L. infantum byla parafyletická u všech použitých algoritmů. MON-30 patří do L. donovani na základě RAPD profilu. A navíc, genetická vzdálenost kmene L. archibaldi a dalších leishmanií nepoukazuje na existencí jednotlivých druhů. (cs)

Fifty-two decameric to 21-meric primers of arbitrary sequence were applied to 15 strains thatbelong to nine zymodemes. Strains belonging to the species L. major and L. tropica were used as outgroups. A total of902 amplicons generated by RAPD were scored. Most primers produced species-specific profiles, only 0.6% ampliconswere shared by all species, while 4.3% amplicons were common for all 15 strains of the L. donovani complex. Wellsupportedtrees have been constructed, which show a rather strong correlation between the genetic polymorphism ofstudied strains and their geographic origin. In all obtained trees, L. infantum was paraphyletic. The RAPD profilessuggest that MON-30 belongs to L. donovani. Moreover, the genetic distance between the L. archibaldi strain and otherleishmanias does not warrant existence of a separate species
