| Description
| - Bovine leukocyte adhesion deficiency (BLAD) in Holstein cattle is a lethal autosomal recessive genetic disease manifested by disturbance of immunity and high mortality in calves (recessive homozygotes) during first months of a life as a consequence of infection complications. The molecular causation of disease is a site mutation at position 383 of the CD 18 gene of chromosome 1. The aim of this study was determination of occurrence from blood of 10 Holstein cows. PCR/RFLP method was used with 10 restriction endonucleases. In 3 from 9 amplificates were identified polymorphisms. Two polymorphisms were identified with Taq I (T/CGA), one polymorphism with Tru I (Mse I) (T/TAA). Presented results indicate the possibility of multiple polymorphisms occurrence in monitored sequences of locus CD18 of chromosome 1 in Holstein cattle. Further, the whole chromosome sequence of CD 18 locus will be analysed.

- Bovinní leukocytární adhezní deficience (BLAD) je letální autozomálně recesivní chorobou, která je manifestována narušením imunity a vysokou mortalitou telat (recesivních homozygotů) v průběhu prvních měsíců života následkem infekčních komplikací. Molekulární příčinou choroby je mutace na pozici 383 genu CD18 na chromozomu 1. Cílem studie je určení výskytu polymorfismu v části lokusu CD18 u dominantních homozygotů. DNA byla izolována z krve 10 krav. Byla provedena PCR/RFLP s 10 restrikčními endonukleázami. 3 z 9 amplifikací identifikovaly polymorfismus. Dva byly nalezeny s využitím Taq I (T/CGA), třetí s Tru I (Mse I) (T/TAA). Výsledky naznačují možnost výskytu četných polymorfismů v monitorované sekvenci lokusu CD 18 na 1BTA. (cs)
