| - 8-hydroxy-2’-deoxyguanosine (8-OH-2’-dG) is one of the predominant forms of radical-induced lesions to DNA, and therefore it has been widely used as a biomarker for oxidative stress. We studied an analytical procedure of determination of urinary 8-OH-2’-dG. A routine assay of free 8-OH-2’-dG in body fluids such as urine is problematic, therefore the purification of this adduct from urine employs solid-phase extraction (SPE) procedure. Various SPE columns from different producers were used as follows: Waters – Oasis MCX (sorbent silica + benzensulfonic acid), Supelco Discovery C18 Oktadecyl (sorbent silica + C18), UCT Columns – Clean Screen DAU206 (sorbent silica + C18 + benzenculfonic acid) and Styre Screen DBX (sorbent styrene-divinylbenzene + C18 + benzensulfonic acid), Macherey-Nagel Chromabond HR-X (sorbent styrene-divinylbenzene) and HR-X-C (sorbent styrene-divinylbenzene + benzensulfonic acid), Phenomenex STRATA X-C (sorbent styrene-divinylbenzene + benzensulfonic acid).
