Description
  • The aim of the study was to examine quality of in vivo-derived cattle embryos. The embryos were recovered from superovulated Holstein cows (n = 59), classified for body condition score (BCS) according to a 5-point scale (BCS2- n = 17; BCS3- n = 33; BCS4- n = 9), on 6th day after the first insemination by a non-surgical flushing of the uterine horns. The embryos were morphologically evaluated for developmental stage (morula, blastocyst, fragmented/degenerated) and analyzed for apoptosis (TUNEL), total cell number (DAPI), actin cytoskeleton quality (phalloidine-TRITC) and cell ultrastructure (transmission electron microscopy). The highest embryo yield (69.3%) was obtained in the BCS3 group, but with the BCS elevation the embryo yield decreased and the last embryo yield (35%) was obtained in the BCS4. The lower embryo cell count and higher number of embryos with cytoplasmic fragmentation were recorded in the BCS4, compared to BCS2 or BCS3 groups. Apoptotic index was highest in the BCS4 embryos compared to the BCS2 group, whilst the BCS3 embryos showed a middle value. The BCS4 group had a twice less proportion of embryos with high quality actin cytoskeleton, and these embryos were characterized by a higher incidence of damaged and vacuolated mitochondria as well as lipid droplets compared to lower BCS groups. These results show that elevation of BCS over an average value may result in worse embryo quality.
