  • Kmín (Carum carvi L.) je tradiční miříkovitou léčivou a cizosprašnou rostlinou. Konvenční šlechtitelské metody se soustředěné na vysoký výnos a kvalitu silice v semeni jsou omezeny nízkou genetickou proměnlivostí (Nemeth 1998, Evenhuis et al. 1998). Současná studia rodu Daucaceae nenásledují možnosti dihaploidních rostlin přes in vitro pylovou embryogenezi kromě mrkve (Andersen et al. 1990, Adamus a Michalik 2003). Pro ověření schopnosti tvorby DH rostlin u kmínu bylo testováno 10 vybraných genotypu ozimého kmínu. Stav ploidie z regenerovaný rostlin byl analyzovaný pomocí flow-cytrometrie. Začátek pylového vývoje z normální gametogeneze k sporogenezi byla zjištován metodami světelné a fluorescenční mikroskopie. Donorové, haploidní, dihaploidní, andiploidní rostliny byly srovnávané analýzou esterasáz (EST) pro rozlišení pylového nebo prašníkového původu regenerujících rostlin. Četnost indukce embryogeneze byla nízká. Mikroskopie metoda ukázala asynchronní vývoj pyl uvnitř prašníků. (cs)

Conventional breeding methods concentrated on high yield and quality of oil compounds of seeds are limited by low genetic variability (Nemeth 1998, Evenhuis et al. 1998). Current studies in Daucaceae family do not follow a possibility of dihaploid plants production trough in vitro pollen embryogenesis except carrot (Andersen et al. 1990, Adamus and Michalik 2003). The 10 selected genotypes of winter type caraway were tested for their ability to produce dihaploid plants (DH). The ploidy status of the regenerated was analysed using flow-cytometry. The beginning of switch pollen development from normal gametophytic to sporophytic pathway was investigated by methods of light and fluorescent microscopy with acetocarmine and DAPI staining, respectively.
