Description
| - Three basic regeneration protocols were used in a set of experiments focused on the development of reliable, routine and genotype-independent transformation method in pea:(1) multiple shoot culture (MSC) derived from axillary meristems, (2) somatic embryogenesis (SE) from apical meristems,(3) trimmed germinating seeds (TGS). The indirect transformation via Agrobacterium tumefaciens (EHA 105) carrying plasmids pBIN19 and pGT89 was used for the basic studies. SE-based Agrobacterium transformation approach resulted in the lowest transformation efficiency (TE)1-2%, TE of MSC and TGS was 18 % and 30 % in T0 generation, respectively. SE regeneration protocol was also used for the experiments with direct gene transfer via particle bombardement with plasmid pDB1. Variants of pressure / grid combinations were studied for optimal particle gun setting. The efficiency of transformation via particle bombardement was 0,5-1,4 %.
